Differential regulation of amphetamine-induced serotonergic and dopaminergic efflux by syntaxin 1A

چکیده
Background The plasma membrane serotonin transporter (SERT) is a key regulator of synaptic serontonergic neurotransmission and is a major target of both antidepressents and psychostimulant drugs of abuse. The pre-synaptic soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) protein syntaxin 1A has been reported to modulate the intrinsic activity of multiple monoamine neurotransmitter transporters both in vitro and ex vivo. However, in contrast to the dopamine transporter (DAT) little is know of its effect on SERT-dependent amphetamine-mediated efflux in neuronal cells. Thus, the purpose of this study was to examine the specific effects of syntaxin 1A on both SERT function and regulation by common drugs of abuse.

Simple transporter trafficking model for amphetamine-induced dopamine efflux.
Amphetamine and its derivatives are important drugs of abuse causing both short-term excitatory and long-term addictive effects. The short-term excitatory effects are linked to amphetamine's ability to maintain high levels of dopamine (DA) outside the cell both by inhibiting DA reuptake after synaptic transmission and by enhancing the efflux of DA from the dopaminergic cells. The molecular mech...
متن کاملThe effects of Adrenergic agents on the dopaminergic-induced sniffing
There is evidence indicating that adrenoceptor mechanisms may influence some of the behaviors in rat. However the role of adrenoceptor agents on sniffing has not been identified. In the present study, the influence of adrenoceptor agents on sniffing induced by apomorphine and amphetamine has been investigated. Male Albino rats, weighing 150-250 g were used for all experiments. The behavior wa...
متن کاملSyntaxin 1A interaction with the dopamine transporter promotes amphetamine-induced dopamine efflux.
The soluble N-ethylmaleimide-sensitive factor attachment protein receptor protein syntaxin 1A (SYN1A) interacts with and regulates the function of transmembrane proteins, including ion channels and neurotransmitter transporters. Here, we define the first 33 amino acids of the N terminus of the dopamine (DA) transporter (DAT) as the site of direct interaction with SYN1A. Amphetamine (AMPH) incre...
متن کاملHydroethanolic extract of Carthamus tinctorius induces antidepressant-like effects: modulation by dopaminergic and serotonergic systems in tail suspension test in mice
Objective(s): Studies indicate that major deficiency in the levels of monoaminergic transmitters is a reason for severe depression. On the other hand, it is shown that Carthamus tinctorius L. (CT) may improve neuropsychological injuries by regulation of the monoamine transporter action. Hence, the present study was undertaken to evaluate the involvement of monoaminergic systems in antidepressan...
متن کاملCalmodulin Kinase II Interacts with the Dopamine Transporter C Terminus to Regulate Amphetamine-Induced Reverse Transport
Efflux of dopamine through the dopamine transporter (DAT) is critical for the psychostimulatory properties of amphetamines, but the underlying mechanism is unclear. Here we show that Ca(2+)/calmodulin-dependent protein kinase II (CaMKII) plays a key role in this efflux. CaMKIIalpha bound to the distal C terminus of DAT and colocalized with DAT in dopaminergic neurons. CaMKIIalpha stimulated dop...
